It mimics part of the body's own protein SNAP-25 and is meant to weaken muscle contraction under the skin - a similar principle to botulinum toxin, but applied on the skin instead of injected.

What exactly is SNAP-8?

SNAP-8 is the further development of the well-known cosmetic peptide Argireline (Acetyl Hexapeptide-3/8). While Argireline consists of six amino acids, SNAP-8 has two additional ones at the N-terminus. The idea behind it: the longer peptide fits its target protein better and inhibits muscle contraction more strongly. It was developed by the Spanish company Lipotec (now part of Lubrizol).

The amino acid sequence is Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH2, and the molecular weight is about 1,075 Da. The CAS number is 868844-74-0. SNAP-8 is water-soluble and is usually sold as a ready-to-use solution (0.05% active peptide in water) or as a lyophilized powder.

How is SNAP-8 used?

SNAP-8 is used only topically - meaning applied to the skin, typically as part of a serum or cream. It is not injected. In the manufacturer-sponsored studies, a 10% solution of the commercial solution (which corresponds to 0.005% pure peptide) was applied to the face twice daily for 28 days [1].

The manufacturer's recommendation is 3-10% of the commercial solution in the final formulation [1][2]. On the gray market, SNAP-8 is occasionally sold as pure powder that you can dissolve in water yourself and then mix into a cream base. This is an unverified user practice without standardized quality control - more on that below.

SNAP-8 is a cosmetic ingredient, not a drug. There is no FDA or EMA approval as an active substance. In the EU and the US, it is listed as a cosmetic ingredient (CosIng reference 54131).

From vial to use: mixing SNAP-8

Storage

SNAP-8 should be stored in the refrigerator at 2-8°C, protected from light and dry. The shelf life of the commercial solution is about 12 months after opening. Lyophilized powder lasts longer when stored properly (typically 24 months). Freeze-thaw cycles should be avoided, as they can damage the peptide structure.

How does SNAP-8 work in the body?

To understand the mechanism, we need to briefly look at how muscles contract in the first place. At the contact point between nerve and muscle (the neuromuscular junction), the nerve releases the messenger acetylcholine. For this, small vesicles carrying the messenger must dock and fuse with the nerve membrane. Docking is mediated by the so-called SNARE complex - a protein scaffold in which the protein SNAP-25 plays a key role.

This is where SNAP-8 comes in. The peptide mimics a section of SNAP-25 and competes with the real protein for its place in the SNARE complex. When SNAP-8 is incorporated instead of SNAP-25, the complex is less stable, less acetylcholine is released, and the muscle contracts more weakly. The result: expression lines caused by repeated muscle contractions are supposed to soften [1][2].

Unlike botulinum toxin, which permanently cuts the SNARE proteins, SNAP-8 is a reversible, competitive inhibitor - it doesn't destroy anything, it only blocks temporarily. This also means the effect fades when you stop using it.

In vitro (i.e., in a test tube), SNAP-8 inhibited glutamate release by up to 43% at a concentration of 1.5 mM [1]. But whether these effects also occur under the skin in living humans is another question - more on that shortly.

What do the studies say?

The study situation for SNAP-8 is thin and mostly manufacturer-sponsored. The most frequently cited study was conducted by Lipotec itself: 17 women applied a 10% SNAP-8 solution twice daily for 28 days. The result was an average reduction in wrinkle depth of 34.98% - compared to 27.05% for Argireline [1]. That sounds impressive, but with 17 participants and no placebo control group, the significance is limited.

An independent, peer-reviewed clinical study on SNAP-8 does not exist. A 2020 review confirms the biochemical mechanism but points out that the clinical data comes almost exclusively from the manufacturer [1]. Another review from 2025 concludes that the evidence for inhibition of synaptic transmission in facial muscles through topical application is 'limited and largely hypothetical' [3].

The penetration problem

This is the central criticism of SNAP-8 and all similar peptides. The skin barrier (the stratum corneum) is designed to keep foreign substances out. The rule of thumb is: molecules over 500 Daltons barely penetrate intact skin. At ~1,075 Da, SNAP-8 is more than double this threshold.

The only available penetration study was not conducted with SNAP-8 itself, but with the related Argireline (~888 Da). After 24 hours on human skin, only 0.22% of the applied peptide was found in the horny layer - and nothing at all in the deeper dermis [3]. Since SNAP-8 is even larger, its penetration is likely equal or worse.

This doesn't necessarily mean SNAP-8 is ineffective - but it's unclear whether enough peptide reaches the neuromuscular junctions to have a measurable effect there. It's possible that such peptides partially work through other mechanisms on the skin surface that aren't yet fully understood.

Safety and side effects

SNAP-8 is considered safe for topical use. The most common side effect is mild, temporary itching at the application site, which stops immediately when washed off. Occasionally, minimal redness may occur [1][3].

However, there are no long-term safety studies beyond 12 weeks. There is also no data for special populations - pregnancy, breastfeeding, under 18, or over 65 years. This is not unusual for a cosmetic ingredient, but it should be kept in mind with longer use.

SNAP-8 vs. Argireline - what's the difference?

SNAP-8 is the further development of Argireline. Both peptides target the same mechanism (SNARE complex inhibition), but SNAP-8 has two additional amino acids that theoretically make it a better docking candidate. In the manufacturer-sponsored comparison study, SNAP-8 performed about 30% better than Argireline [1]. However, since this data comes from the same manufacturer that sells both peptides, it's only of limited significance.
